Identification of H5N1 subtype avian influenza virus from captive Panthera Tigris and analysis of HA and NA gene

Abstract: Influenza A viruses belongs to the genus Strains of Sweat Virus,which is threatening the
health of humans and animals,resulting in plenty of economic loss and increasing the burden on
society.In this study,the tissue samples of died Panther tigris were collected and grinded in PBS.
After centrifugation,the supernatant was added with double antibody and inoculated in 9-10days
 of age chicken embryo,the virus were isolated.The experiments of HA and HI showed that the virus was H5 subtype avian influenza virus;the gene sequence analysis of HA and NA showed that the virus was H5N1 subtype avian influenza virus,the HA gene belongs to the branch of Clade2.3.2.1.The result indicated that the Panther tigers was infected by H5N1 subtype highly pathogenic influenza.This study is significant for controlling the influenza disease of captive panthera tigers.
